A study to assess the biological activity of TIMP-1 in blocking collagenase-induced hydrolysis of [125I]-collagen*
Enzyme Protease Inhibitor Hydrolysis Products (cpm)

No enzyme{dagger} ... 275 ± 28
Collagenase, 10 µg (total counts){ddagger} ... 5750 ± 250
Collagenase, 0.5 µg ... 3470 ± 180
Collagenase, 0.5 µg STI, 10 µg 3320 ± 220
Collagenase, 0.5 µg TIMP-1, 5 µg 480 ± 33
Collagenase, 1.5 µg TIMP-1, 5 µg 760 ± 46

*Assays were performed as described in "Materials and Methods," using highly purified TIMP-1 isolated from SCCM. Each number represents the mean ± SD of triplicate wells from an experiment. This experiment was repeated 3 times using different batches of TIMP-1, and similar results were obtained each time. Data were not pooled since the amount of [125I]-collagen coated per well in each experiment varied, yielding different total counts.
{dagger}Nonspecific control represent counts, ie, [125I]-collagen, that were released nonspecifically from coated wells.
{ddagger}Total counts represents hydrolyzed fragments of [125I]-collagen caused by collagenase-induced proteolysis.
